I n m a n y freeze dryers the embedding process takes place within the freeze dryer without the v a c u u m being broken. T h e paraffin wax must be placed in the dryer before the tissue and degassed under vacuum (see Method 11). This technique, while admirable in m a n y respects, has the disadvantage, in the author's experience, of occasionally causing damage to the tissue.

Water cannot be used, and removing the creases is also a problem.
